毛细管电泳法测定焦山楂中的丙烯酰胺

Determination of Acrylamide in Coking Hawthorn by Capillary Electrophoresis

摘要:

采用毛细管电泳法对高温炮制中草药的丙烯酰胺进行痕量检测.以水提取焦山楂中丙烯酰胺,固相萃取柱净化浓缩,毛细管电泳分离,二极管阵列检测器测定其含量.以33mmol/L磷酸二氢钾-33mmol/L磷酸氢二钠(体积比1∶1)-50mmol/L SDS(pH=6.7)为电泳缓冲液,采用压力进样方式,在10kV恒压下进行分离,并在波长198nm处检测.丙烯酰胺在0.50~50.00mg/L内与峰面积呈良好线性关系,相关系数为0.999 5,检出限为0.10mg/L,回收率在93.3%~98.9%,相对标准偏差小于2.57%.

Abstract:

To study a method of determination for acrylamide in preparing Chinese medicine by capillaryelectrophoresis.The acrylamide in coking hawthorn was extracted by water.The extracting solution waspurified and concentrated with solid phase extraction column,then detected by photodiode array dection.The solution consisting of potassium dihydrogen phosphate(33mmol/L)disodium hydrogen phosphate(33 mmol/L)with a volume fraction of 1∶1,and SDS(50mmol/L,pH=6.7)was used as the running buffer.The applied voltage was 10kV and detection wavelength was set at 198nm.The calibration curve was line-ar with peak area in a range from 0.50mg/L to 50.00mg/L,and the linear relativity was 0.999 5.The de-tection limit of the method was 0.10mg/L,and the recovery was 93.3%~98.9%,with RSD lessthan 2.57%.
