A Method to Detect Phoxim Residue by Analyzing the Inhibition of Cholinesterase

摘要:

利用胆碱酯酶水解碘化硫代丁酰胆碱后形成的硫代胆碱,与二硫双对硝基苯甲酸发生化学反应生成黄色产物,在 412nm比色测定.利用此法测定酶、底物的添加量以及保温温度和保温时间对酶活力的影响,还研究了不同浓度与保温时间的辛硫磷对胆碱酯酶活力的抑制情况.获得了胆碱酯酶优化的反应条件,结果还表明胆碱酯酶对辛硫磷比较敏感,其抑制中浓度为I50 为 3.90 49μg/L.

Abstract:

Ellman reaction is a simple and sensitive method to analyze cholinesterase activity. A new found cholinesterase was studied to get the best reaction condition through Ellman reaction, and it was found this cholinesterase was very sensitive to phoxim, its medium inhibition concentration I50 to this cholinesterase was only 3.904 9mg/L, so it had good promise to be used in detecting organophosphate and carbamate pesticide residue in agricultural products.
