Selection of the Mutation Strain with High Productivity by Protoplast Fusion Technique

摘要:

以龟裂链霉菌(Sterpomyces rimosus)98#和76#为出发菌株,分别用溶菌酶制得2个亲本的原生质体,其中76#采用15 W紫外灯下30 cm照射25 min,100%灭活,用PEG诱导进行了原生质体的融合。通过比较菌落外观、颜色挑选融合子328株,结合前期代谢情况进一步筛选高单位的融合菌株8株,经反复传代考察融合子的稳定性后得到1株新菌株102#,其发酵单位比亲本提高了13。0%。

Abstract:

The protoplasts of strains 98# and 76# of Sterpomyces rimosus were obtained by dissolvent enzyme respectively.The protoplasts of strains 76# were treated with UV light for 15 W,30 cm and 25 min,the death rate reached 100 %.The protoplasts fusion was induced by PEG.328 strains were obtained by selecting the fusion-stains with the character and color of colony,then 8 strains were obtained by further selecting with metabolism,the stability of more progeny was tested and at last one strains 102# was obtained,the ferment level was improved 13.0 % than parent strains.
