稳定转染Oct-4基因后人脂肪来源干细胞性状分析

摘要:

观察了转染Oct-4基因对人脂肪来源干细胞性状的影响.首先将Oct-4基因全序列亚克隆至逆转录病毒载体pMSCVneo中,构建逆转录病毒重组质粒,然后以脂质体介导转染包装细胞PT67,成功建立PT67-Oct4细胞系,随后转染人脂肪来源干细胞(adipose-derived stemcells,ADSCs)中,最后对高表达Oct-4基因的ADSCs性状进行观察.实验结果表明,对高表达Oct-4蛋白的ADSCs-Oct4同未转染Oct-4的ADSCs相比,ADSCs-Oct4在形态学、表面标志、核型等方面无明显差别,接种ADSCs-Oct4细胞8周后裸鼠体内未形成肿瘤.以上结果说明ADSCs-Oct4细胞遗传性状稳定,无致瘤性,为深入开展ADSCs研究提供了实验依据.

Abstract:

The effects of Oct-4 gene transfection on human adipose-derived stem cells (ADSCs) were observed.At first,the whole sequence of Oct-4 gene was sub-cloned into retroviral vector pMSCVneo to construct the recombined ret rovirus vector pMSCV-Oct4,and then the recombined plasmid was transferred into packaging cell line PT67 with lipofectamine.Finally PT-Oct4 cell lines were successfully established. ADSCs were transfected and observed.The results showed that the ADSCs-Oct4 had no obvious dif ference in morphology,surface marker and karyotype compared to unt ransfected ADSCs.Furthermore,there was no tumor formed in nude mice 8 weeks after ADSCs-Oct4 inoculation. The above results revealed that the ADSCs-Oct4 had stable genetict raits,no tumorigenicity,and provided the experimental basis for in-depth research.
