环鸟苷酸信使系统参与rbcS基因表达调控初报

摘要/Abstract

摘要:

以转 rbc S-GUS基因烟草悬浮细胞为材料, 通过用荧光光度法检测报告基因 GUS酶的活性的方法, 发现暗中培养的转基因烟草悬浮细胞经细胞外 Ca M或红光处理后, rbc S基因表达明显增加 (分别增加 2~ 3倍和 7倍 ), 而细胞外 Ca M和红光同时处理却只使 rbc S基因表达增加 4~ 5倍.在暗中培养的转基因烟草悬浮细胞介质中加入 8-Br-c GMP使 rbc S基因表达降低 50 %左右, 而加入鸟苷酸环化酶拮抗剂 LY83 583和 Genestin则使 rbc S基因表达增加约 50 %.上述结果表明, 细胞外 Ca M和红光在调控 rbc S基因表达时存在拮抗关系, 并且 c GMP信使系统可能参与了 rbc S基因表达的抑制调控

Abstract:

We used transgenic tobacco with rbcS-GUS fusion gene as experimental materials and examined the expression of rbcS-GUS gene through the activity analysis of GUS. Adding purified CaM to suspension-cultured tobacco cells resulted in an increase of the expression of rbcS about 2 ~ 3 folds higher than that of control and red light illumination resulted in 7 folds higher. But purified CaM together with red light treatment only increased the expression of rbcS about 4 ~ 5 folds. The expression of rbcS decreased 50 % after adding 8-Br-cGMP to the suspension-cultured cells in dark, while adding LY83583 and Genestin increased the expression of rbcS about 50 % . These data showed extracellular CaM and red light signaling pathways may inhibit each other, and cGMP signaling pathway may involve in the repression of rbcS gene.
