利用DDRT-PCR技术分离和克隆玉米抗大斑病相关基因片段

Identification of Expressed Gene Fragments Related to Corn Resistance to Setosphaeria turcica by DDRT-PCR

摘要:

分离和克隆农作物的抗病基因是植物病理学及分子生物学研究的热点领域,笔者以一套含有不同主效抗病基因的玉米自交系B37,B37Ht1,B37Ht2为试验材料,接种玉米大斑病菌0号小种菌株01-27后,利用mRNA差异显示反转录PCR(mRNAdifferentialdisplayreversetranscriptionPCR,DDRTPCR)技术分离玉米抗大斑病相关基因片段.通过反式Northern杂交检测,共获得了44条差异表达基因片段,其中与B37亲和性互作相关的片段17条,与B37Ht1和B37Ht2非亲和性互作相关的片段分别为11条和8条.

Abstract:

Identification and cloning the disease-resistant genes of crop is one of the most important research fields to plant pathologists and molecular biologists. In the present study, expressed gene fragments related to corn resistant to Setosphaeria turcica have been identified from corn inbred lines B37, B37Ht1 and B37Ht2 by the mRNA differential display reverse transcription PCR ( DDRT-PCR). After inoculated with isolate 01-27 belonging to race 0 of S. turcica, forty-four differentially expressed gene fragments have been obtained after being detected by reverse Northern blotting. Out of them, seventeen fragments were found to be related to compatible interaction between corn inbred line B37 and isolate 01-27, eleven fragments related to incompatible interaction between B37Ht 1 and the isolate, and eight fragments related to incompatible interaction between B37Ht2 and the isolate.
