鲟鱼海豚链球菌的分离鉴定

Isolation and Identification of Streptococcus iniae in Sturgeon

摘要:

鲟鱼是重要的冷水鱼养殖种类之一,但在养殖过程中,链球菌病的频繁暴发对鲟鱼幼鱼及鱼苗的健康造成了重大威胁,严重者导致极高的死亡率.2023年8月,河北省平山县多家冷水鱼养殖场出现大量鲟鱼死亡病例.收集了被感染的鲟鱼病例,通过临床症状观察、形态学分析、分子鉴定及药敏试验等,对被感染的鲟鱼进行诊断.细菌分离培养结果显示,从发病鱼肝脏和血液中分离出了链状革兰氏阳性菌,初步确定为链球菌.16S rRNA序列比对分析结果为海豚链球菌.药敏试验结果显示,分离菌株对头孢氨苄、大观霉素和环丙沙星等7种药物敏感.研究结果为河北省平山县冷水鱼链球菌病的防治提供了参考依据.

Abstract:

Sturgeon is one of the most important species of cold-water fish cultured. However,during the farming process,frequent outbreaks of streptococcosis pose a significant threat to the health of sturgeon juveniles and fry,resulting in extremely high mortality rates in severe cases.In August 2023,a large number of sturgeon deaths occurred at several cold-water fish farms in Pingshan county,Hebei province. In this study,the above infected sturgeon cases were collected,and the diagnosis of the cases were carried out by clinical symptom observation,morphological analysis,molecular identification and antimicrobial susceptibility testing.The results of bacterial isolation and culture demonstrated that the gram-positive bacteria arranged in chain were isolated from the liver and blood of the diseased fish,and they were initially identified as Streptococcus.The results of 16S rRNA sequence alignment analysis showed that the isolates were Streptococcus iniae.Antimicrobial susceptibility testing revealed that the isolated strains were sensitive to 7 drugs,such as cephalexin,spectinomycin and ciprofloxacin.This study provides a reference for the prevention and treatment of cold-water fish streptococcosis in Pingshan county,Hebei province.
