Construction of Escherichia coli-mycobacterium Shuttle Expression Plasmid pY -α
摘要/Abstract
摘要:
利用分子生物学方法,以含人结核杆菌热休克蛋白(HSP)70基因全长序列的质粒pMT70为模板,扩增出hsp70启动子,将其定向克隆于pUC19,构建成重组质粒pY6013;然后以重组质粒pIJK1为模板,扩增出分枝杆菌α信号肽基因并将其克隆到pY6013质粒中hsp70启动子的下游,得到了大肠杆菌分枝杆菌表达质粒pY-α.
Abstract:
With cloning technique used,firstly,human Mycobacterium tuberculosis heat shock pro-tein( hsp) 70 promoter in plasmid pM T-70 was amplified by PCR. Hsp 70 promoter was cloned into pUC19 and the pY6013 was constructed. Secondly the gene for the crossing-reactive α antigen of Mycobacterium kansasii in plasmid pIJK-1was amplified and cloned into plasmid pY6013. Then a new E coli - mycobacterium shuttle expression plasmid pY- αwas constructed.
