Study of miRNA's Influence in the Pathological Typing of Breast Cancer Based on miRNA Regulatory Networks

摘要:

目前microRNA(miRNA)转录后的调控作用在乳腺癌(breast cancer,BC)致病机理探寻中受到广泛关注.但在病理分型中,人们对于miRNA如何与其靶标基因相互作用来调控乳腺癌的分型还知之甚少.本文通过结合基因表达数据和序列信息预测miRNA在乳腺癌分型中的作用.首先,使用T统计针对健康、非浸润及浸润性乳腺癌样本提取差异表达的miRNA和mRNA;然后,利用HCtarget算法预测表达显著miRNA的靶基因;最后,构建miRNA和mRNA调控网络并进行富集性分析.分子生物学分析显示,10个显著表达的miRNA的靶基因广泛参与MAPK信号通路、Wnt信号通路、雌激素信号通路等与乳腺癌密切相关的通路.而hsa-miR-200c,hsa-miR-205和hsa-miR-145调控强度变化贯穿乳腺细胞由正常到非浸润再到浸润的整个过程,可以作为识别乳腺癌潜在的分子标志物,为乳腺癌发病机理研究提供了方法和依据.

Abstract:

At present,the post-transcriptional regulation of microRNA (miRNA) in the exploration of breast cancer (BC) has attracted widespread attention.However,in pathological typing,it remains poorly understood that how miRNA interact with their target genes to regulate breast cancer typing.The effect of miRNA in the classification of breast cancer is predicted in this article through integrating expression data and sequence information.First,we used T statistics to extract differentially expressed miRNA and mRNA from healthy,non-invasive and invasive breast cancer samples;then we used HCtarget algorithm to predict the target genes of significantly expressed miRNA in breast cancer samples;at last,we constructed a regulatory network of miRNA and mRNA and did enrichment analysis.The molecular biology analysis showed that 10 significantly expressed miRNA and their target genes extensively participated in the MAPK pathway,Wnt pathway,the ER signaling pathway and other pathways that are closely associated with breast cancer.Additionally,the changes of regulatory intensity of hsa-miR-200c,hsa-miR-205 and hsa-miR-145 existed in the whole process of breast cells from normal to non-invasive and to invasive state,therefore,they can be used as potential molecular markers for identification of breast cancer.This article has also provided a novel approach and basis for studying the pathogenesis of breast cancer.
