摘要
利用控制酶的固定量和使用外层膜等手段,限制底物葡萄糖的扩散,制备了扩大响应线性范围的葡萄糖氧化酶电极.考察了酶电极上酶的不同固定量及外层膜组成比对线性响应的影响.结果显示:葡萄糖氧化酶的固定量20 μg,外层膜组成比V(TMOS)∶V(H2O)∶V(甲醇)=0.1∶0.1∶0.5时,制备的酶电极对葡萄糖响应的线性范围为8.0×10-5~1.2×10-2 mol/L,线性范围扩大了2~3倍.

Abstract:
An approach is described to extend the response linear range of glucose oxidase electrode prepared by the control of enzyme loading and the use of external membranes to restrict diffusion.Effects on the linearity of enzyme electrode prepared have been studied by changing enzyme loading and constitute of the outer layer.The results show the approach has given linearity up to 12.0 mmol/L glucose with the enzyme loading of 20 μg and V(TMOS)∶V(H2O)∶V(methanol) is 0.1∶0.1∶0.5 in the outer membrane prepared,while extending 2 to 3 times as wide as linear range of the enzyme electrode without the outer membrane.
