摘要
通过响应面法优化木藤蓼多糖提取工艺,采用苯酚硫酸法测定木藤蓼中多糖含量.实验考察了温度、料液质量浓度、超声时间等单因素对多糖提取的影响;在单因素的基础上,采用BOX Behnken进行3因素3水平中心组合设计并进行实验.结果显示,在温度65℃、质量浓度450mL/g、超声40min条件下,木藤蓼多糖含量达到最大值,为0.59%.实验的稳定性、重复性、精密度的相对标准偏差分别为0.87%,0.84%,1.09%,实验方法可靠.

Abstract:
The extraction process of polysaccharides from Fallopia aubertii L.Henry Holub by response surface methodology was optimized,and the polysaccharide content of Fallopia aubertii L.Henry Holub was determined by phenol sulfuric acid method.The effects of temperature,material-liquid ratio and ultrasonic time on the extraction of polysaccharides were investigated experimentally.On the basis of single factor,BOX Behnken was used to design a three-factor three-level center combination experiment.The results showed that the polysaccharide content of Fallopia aubertii L.Henry Holub reached a maximum of 0.73% at the temperature of 65℃,the material mass concentration of 450mL/g,and the ultrasonic time of 40 min.The relative standard deviations of stability,repeatability and precision of the experiment are 0.87%,0.84% and 1.09%,showing that the experimental method is reliable.
